How can this medicine affect other medicines?
Amprenavir should not be taken with the following due to a risk of serious or life threatening side effects:-
- terfenadine
- cisapride
- pimozide
- the benzodiazepines triazolam, diazepam, flurazepam and midazolam
- ergotamine
- dihydroergotamine
The following should not be taken with amprenavir as they can make it ineffective:-
- rifampicin
- the herbal remedy St John's wort
Ritonavir increases and efavirenz and nevirapine may reduce the blood levels of amprenavir. Dosage adjustments may be made by your doctor if ritonavir, nevirapine or efavirenz are part of your anti-HIV treatment.
Delavirdine may increase the blood levels of amprenavir.
Amprenavir increases the blood levels of rifabutin, therefore lower than normal doses of rifabutin may be prescribed.
Doses of antacids or indinavir should be separated from amprenavir doses by at least an hour, as they may decrease the absorption of amprenavir from the gut.
Amprenavir has the potential to interact with many other medicines, but as it is a new medicine, there is not yet sufficient information to determine how important these interactions might be. If the following are taken with amprenavir, they should be monitored by your doctor:-
- opiate derivatives such as methadone
- amiodarone
- phenobarbitone, phenytoin
- tricyclic antidepressants
- quinidine
- warfarin
- the calcium channel blockers diltiazem, nicardipine, nifedipine or nimodipine
- sildenafil (Viagra), whose blood levels may be increased
- cholesterol lowering medicines atorvastatin, cerivastatin, fluvastatin, lovastatin, pravastatin and simvastatin
- clozapine, carbamazepine, erythromycin, cimetidine, dapsone, itraconazole, loratidine and benzodiazepines, whose blood levels may also be increased by amprenavir
- hormonal contraceptives, which may have a reduced contraceptive effect. A barrier method of contraception should be used by women of child bearing potential.
